Semaglutide is a GLP-1 receptor agonist sold as Ozempic, Wegovy and Rybelsus, approved for type 2 diabetes and obesity and now among the most-used drugs on the market. The research pool around it has widened well past blood sugar and appetite, and the honest summary is that the most eye-catching findings are also the furthest from human evidence.
Aging markers in old mice
A National Institutes of Health funded study at UC Berkeley gave semaglutide to 20-month-old female mice for three months and reported better muscle and cognitive function, reduced inflammation, and less decline in tissue repair capacity compared with untreated animals. In a separate group treated until death, median lifespan was nearly 100 days longer than in untreated mice. This work was done entirely in mice. The researchers also ran a head-to-head against a 24% calorie-restricted diet and reported that the treated animals held their metabolic rate steady while the calorie-restricted ones slowed, which the authors say hints at a pathway independent of eating less.
A genetic link to male-pattern hair loss
Researchers at NYU Langone compared genetic patterns tied to GLP1R activity with genes found in men who have androgenetic alopecia, and reported an additional 7 percent risk that persisted after adjusting for hypertension, insulin resistance and reduced testosterone. Because male hair loss has been studied far more than female, the analysis covered men only. The databases used were drawn from mostly White participants.
Symptoms patients raise that trials miss
A University of Pennsylvania team analyzed more than 400,000 Reddit posts from nearly 70,000 users of semaglutide and tirzepatide, published in Nature Health. About 44% described at least one side effect, most commonly gastrointestinal. Nearly 4% of those reporting side effects described reproductive symptoms including irregular or heavy menstrual bleeding, and body temperature complaints such as chills and hot flashes also recurred. The authors are explicit that this shows association in what people discuss online, not causation.
The counterfeit problem
Australia's TGA issued fresh warnings after a patient was hospitalized with a torn oesophagus following use of a product sold as retatrutide that testing found contained undeclared semaglutide at eight times the level found in products on the Australian Register of Therapeutic Goods. TGA Chief Medical Advisor Professor Robyn Langham said the biggest risk with unapproved peptide products is not knowing what is in the vial.
What Isn't Established
The lifespan and aging results have not been shown in people, and the authors say clinical research is needed to determine whether any of it translates. The hair loss finding is a genetic association, not a trial outcome, and its senior investigator frames screening and preventive co-treatment as things that depend on future experiments succeeding. The Reddit analysis draws on users who skew younger, more male and US-based, and its authors state plainly that they cannot say GLP-1 drugs cause the symptoms described.
Where the evidence sits
Semaglutide is unusually well studied for a peptide drug, and that is exactly why the gaps are visible. The longevity work is preclinical, the hair loss work is genetic inference, the side-effect signals are observational, and the safety story with the clearest human casualties so far involves counterfeit vials rather than the approved product.
